RAPD-Based Identification and Cluster Analysis of Soybean Cultivars (Glycine max L.) in Henan

Abstract: Ten soybean cultivars'total DNAs were extracted by CTAB method and used for identification and cluster analysis based on RAPD method.Out of 73 random primers,12 were selected to generate stable and polymorphic DNA,fragments.Total 67 RAPD fragments were amplified,51 of which were polymorphic,and whose size varied from 0.2 to 5 kb.Each primer could generate 4 to 9 DNA fragments.The percentage of polymorphic loci was 76.16 %.The results analyzed by SPSS11.0 version software showed that Jaccard'similarity coefficients ranged from 0.528 to 0.776,whose mean was 0.6416,indicating that the genetic variations existed among the 10 soybean cultivars.A dendrogram established based on Hierarchical cluster analysis could divid 10 soybean cultivars into 3 groups,among which there were some genetic relations.
