Abstract: Some primary factors causing hard and slow rooting and high abnormal root percentage in tissue culture of Asparagus officinalis L.were studied. Results indicated that rooting induction was closely related to the heterogeneity of materials, the growth state of explant, growth regulators and their concentrations, and generations of subculture, etc.In a same variety, rooting directly from apexes showed a higher rate than that from callus.A similar trend was observed in the case of normal root percent. The healthy bud lengthening 2-3cm is optimum for inducing, while a weak one needs rejuvenating before induction. The medium suitable for rooting is 1/2MS+VB10.3mg/L+IBA 2.5mg/L+BA 0.25mg/L, which results in rooting rate as high as 86% (the mean root number 9.26) and reduces the abnormal root to 8%.It is best that the subculture is confined within 16 generations.
