Identification of Pathogenic Nematode Isolated from Foxtail Millet in Hebei Province

Abstract: To identify a nematode isolate from infected ears of millet in Hebei province,the morphological characteristics and measurements of the nematode isolate were acquired and compared,its 28S rRNA-D2/D3 and ITS regions of ribosomal RNA were amplified,sequenced and aligned,and the phylogenetic trees were constructed by Neighbor-joining method using MEGA6.0 software.The results showed:this population was very similar to other Aphelenchoides besseyi populations reported before;compared with the sequences from GenBank,the 28S rRNA-D2/D3 regions of the nematode isolate was 100% similar to Aphelenchoides besseyi and the similarity of ITS was 98%-99%.The NJ trees showed that the nematode isolate was clustered into one branch with Aphelenchoides besseyi with 100% support rate.All the information revealed the nematode isolate from infected ears of millet in Hebei province should be Aphelenchoides besseyi.
