Genetic Differentiation among Isolates of Plasmodiophora brassicae (Clubroot) from China

Abstract: In order to develop a method for genetic differentiation of Plasmodiophora brassicae isolates with host and geographical origin among Chinese isolates,this study analyzed genetic variations of 43 Chinese isolates of P.brassicae from 12 hosts and 11 Provinces.Gene phylogenetic analysis of the isolates was based on actin and polyubiquitin genes,and results showed two divergent clusters Ⅰ and Ⅱ.Based on actin genes,the strains of cluster Ⅰ were collected mainly from middle and lower streams of the Yellow River and South China Beijing,Sichuan,Shandong,Henan,Heilongjiang,Shanghai,Shanxi,Hainan,Yunnan and Xizang Provinces,whereas strains of cluster Ⅱ were collected mainly from Northeast China Liaoning,Jilin,Hubei,Yunnan and Xizang Provinces.Based on polyubiquitin genes,the strains of cluster Ⅰ and cluster Ⅱ were collected from North Liaoning,Shandong,Shanxi,Jilin,Henan,Hainan,Yunnan and Xizang Provinces,and cluster Ⅱ were collected from Sichuan,Hubei,Beijing,Shanghai,Heilongjiang,Yunnan and Xizang Provinces South China,respectively.Besides,isolates from Yunnan and Xizang Provinces were distributed amongst two clusters.Furthermore,strains from the same host were distributed in different genetic groups,which suggested that genetic diversity of isolates of P.brassicae had nothing to do with host species.
