Optimization of Fermentation Conditions of Antagonistic Bacteria F1 Against Muskmelon Fusarium Wilt by Response Surface Methodology

Abstract: In order to find out the optimization of fermentation conditions of antagonistic bacteria F1 against muskmelon Fusarium wilt.Plackett-Burman design was employed to investigate the main factors affecting colony-forming units of strain F1 and confirmed that temperature,content of inorganic salt and bean powder were the main factors.After steepest ascent experiment,response surface analysis and mathematic modeling we obtained the optimum conditions of fermentation as following:4% of inoculation;initial pH value 7;200 r/min of rotating speed;the content of corn powder,bean powder,MgSO4,NaH2PO4,CaCl2 2H2O and K2HPO4 was 30,17.92,0.604,1.51,0.302,1.51 g/L respectively;30.0 ℃ of cultivation temperature.So the colony-forming units of strain F1 increased nearly 117 times to 1.507×109 cfu/mL and the experimental values was 101.0% times to the predict values.These results revealed that the predicted model was reliable and available for the optimization of fermentation conditions of F1 during production.
