Principal Component Analysis and Comprehensive Evaluation on Biochemical Characteristics of Cold Tolerance at Booting Stage in the Near-isogenic Lines from Japonica Rice Lijiangxintuanheigu

Abstract: The biochemical evaluation system were established for screening and identifying species with cold tolerance,105 rice materials,including three standard varieties of strong cold resistance from Yunnan Province,the NILs BC4F8 and BC4F9 generation derived from parents japonica cultivars Lijiangxintuanheigu(donor parent) and Towada(recurrent parent),and their parents,were employed for evaluation of cold tolerance and screening of cold resistance index at booting stage.26 biochemical traits including leaves,anthers,and grains of tested materials were determined under cold water-stress and normal conditions in 2011,2012.Comprehensive assessment of cold resistance based on the principal component analysis,subordinate function values analysis,regression analysis and K-means clustering analysis.The results showed Chlorophyll a,Chlorophyll b,total Chlorophyll,Chlorophyll c content,peroxidase(POD) activity,free amino acid content of flag leaves;soluble protein,soluble sugar,malondiadehyde(MDA) content,catalase(CAT) activity,ascorbic acid(ASA),proline content(Pro) of flag leaves and anthers;soluble protein,soluble sugar,protein,γ-aminobutyric acid(GABA) content of grains significantly influenced the cold resistance.Thus could be used as comprehensive index for identification of cold resistance at the booting stage.The values cold resistance comprehensive evaluation(D) of biochemical traits including leaves,anthers,and grains of tested materials based on the subordinate function values was chosen as object under cold stress condition,through K-means clustering analysis,105 lines of population should be divided into 3 categories including stongly cold-tolerant lines 32,mediate cold tolerance lines 10,and cold-sensitive lines 63.
