Preliminary Research on Pathogenic Secretions Generated by Ditylenchus destructor Thorne

Abstract: To explore Ditylenchus destructor Thorne pathogenic mechanism, a preliminary study was conducted withprotein secretions of D. destructor. After nematode glands growth was stimulated by 5 -methoxy -N, N -dimethyltryptamineand dimethyl sulfoxide, nemade was grinded on ice and secretory granules were acquired. The proteins from the granuleswere abstracted by TCA/Acetone and ammonium sulfate precipitation method. Then pathogenicity of protein was testedwith sweet potato slices.The results showed that the secretions from nematode esophageal gland, especially proteins obv- iously played a pathogenic role. Crude proteins abstracted by ammonium sulfate precipitation had the stronger pathogenicitythan that from TCA/acetone method. And the pathogenic crude proteins were heat sensitive and ultraviolet radiation sens- itive, they almost completely lost pathogenicity at 100? for 30min withwater bath and under 40w, 50 cm UV light expo -sure for 60 min.
