Construction of Ammopiptanthus mongolicus cDNA Library and the Primary Analysis of Expressed Sequence Tags (EST)

Abstract: In order to analyse molecular mechanism of abiotic stress and clone its resistance gene, a cDNA library of Ammopiptanthus mongolicus winter leaf was constructed with pBluescript II SK (+) vector. The library consisted of 2.16×105 cfu with the recombinant percentage of 89.6%. Average length of inserted fragment was over 1 kb. Total 542ESTs were obtained with randomly sequenced then 360 uniESTs were obtained by contiganalsis. The analysis of the 360 uniESTs through BlastN and BlastX searching on line showed that 313 uniESTs were determined to be the tags of genes with putative function, these genes were found to be involved in the biological processes including energy metabolism, material transform ation and cell growth. Among these uniESTs, 48uniESTs were determined to be the tags of genes related to stress tolerance.
