Fas Pathway in Apoptosis of BMM Induced by Testosterone

Abstract: In this paper,mechanism involved in suppression of testosterone on immune system through investigating apoptosis of BMMs(bone marrow-derived macrophages) induced by testosterone and Fas signal pathway were discussed.After BMMs were treated with various concentration of testosterone(1-10nmol/L) for 24h,the apoptosis was analyzed by DNA fragment electrophoresisand flow cytometry with Annexin-V and PI double staining methods,and the effect of testosteroneon expression of some genes involved in Fas pathway was estimated by RT-PCR semi-quantified method.BMMs treated with 10 nmol/L testosterone had significantly higher apoptosis ratio 31.7% than contrast BMMs 7.4%,accompanying with the increasing transcription of related genes in Fas pathway such as Fas,FasL,FADD,caspase-8 and caspase-3.Then testosterone might induce apoptosis of BMM through Fas pathway to exert immunosuppression.
