Selection of New Germplasm Resource Using the Technique of Pollen Tube Pathway Transfer Exogenous DNA into Recipients of Oats

Abstract: Using a series of naked oat variety as recipients, such as early-ripe PIN2 mid-late maturing Jizhangyou 4 and late-maturing Jizhangyou 5 and varied-ploidy oats, peanut, soybean and wheat, etc. as the donor, Exogenous DNA was transfered into recipients by pollen tube method, and variations had been obtained. After selected and identified generation-by-generation a series of germplasm resource of naked oats were developed. In order to further test the efficiency of transformation by pollen tube method GUS report gene was introduced as exogenous target gene into oats. In the colony of GT1,a great number of positive plants were inspected what expressed the activity of GUS gene.
