High Efficiency Regeneration and Transformation of Lettuce (Lactuca sativa var. capitata L.)Using Agrobacterium Vector

Abstract: An efficient procedure for obtaining transgenic lettuce plants using agrobacterium is described. The young leaf sections,when cultured on MS medium with 0.5mg/L BA,0.1 mg/L NAA,had a 87% plant regeneration rate, and more than one shoot were regenerated per regenerable explant during culturing. After co-cultured will a}muacteria,the explants had a 65% regeneration rate on MS medium with 100mg/L kanamycin. These regenerated plants showed a 50% positive reaction frequency when their leaf slices incubated with X-Glu solution.
