Selection of Salt Tolerant Variants from Wheat Anther Callus Culture

Abstract: By use of anther culture technique,salt-tolcrantc variants of winter wheat were screened at the cell level by adding quantitative NaCl into the medium C17.About 143331 anther of F1 were cultured on the medium by direct screening and incrcasing salinity screening method.The results proved that the later was a more effective measure for obtaining salt tolerante variants.Green differentiation frequencies on C17+0.1%,NaCl-(callus)-C17+0.3% NaCl and C17+0.1 % NaCl-callus-C17+0.5% NaCl increased 5.06 and 15.0 times respectively,compared with direct screening method,by calculating the number of anthers cultured.Pollen plants differentiated on higher salinity medium C17(added 0.3% and 0.5% NaCl)and their progeny(H2 H1)have higher salt-tolerance.They could develop normally or basically normally in the soil containing 0.4% salimity.
