摘要: 以玉米DNA拓扑异构酶Ⅰ基因Top1的克隆为例,发展了获得玉米基因组全长cDNA序列的3种辅助策略:逐步展示法、保守结构域策略和BAC途径,并对这3种策略的效用进行了评价。

Abstract: Take maize DNA Topoisomerase N gene Top1 cloning as an example, three accessorial methods developed for achieving full length cDNA sequence in maize genome, that were step by step approach, conserved domain strategy and BAC technique, were described. The three accessorial methods were also assessed primarily in this study.
