苹果实生树韧皮部中阶段转变特异蛋白质的SDS-PAGE分析

SDI PAGE Analysis of Phase Change- related Proteins in Phloem in Apple( Malus domestics Borhk.)

摘要: 利用聚丙烯酰胺凝胶电泳技术(SDS-PAGE)分析了5株苹果杂种实生树(红玉×金冠)韧皮部中蛋白质含量随节位升高的动态变化。结果表明,随节位升高蛋白质带发生变化,共发现3条阶段转变特异蛋白质带,分子量估算值分别为17.0,21.0,55.0 kDa。其中55.0 kDa蛋白质表现为定性变化,从46节出现,之后含量保持稳定,到125节后突然消失。21.0 kDa蛋白质111~125节以上含量突然降低 17.0 kDa蛋白质在在实生树低节位含量低,在51~65节以上含量突然增加并保持稳定,到106~120节含量突然下降。

Abstract: Dyanic changes of proteins in phloem from different nodes in 5 hybrid seedlings( Jonathan x Golden De-licious)were analyzed with SDI polyac amide gel electrophoresis. The result indicated that protein bands changed withnodes increasing.Three prater bands found in different phases. The molecular weight of the 3 protein bands were17. 0, 21. 0, 55. 0 kDa respectivly.The 55. 0 kDa protein presented qualitative change which appeared in the node No.46 and disappeared the node No. 125. Changes of 17. 0 kDa and 21. 0 kDa proteins were found quantitaively. In phloemsamples taken below nodes No. 111-125, the content of 21. 0 kDa protein was higher, but a sharp decrease occurred inphloem above nodes No. 125. The content of 17. 0 kDa protein kept high bet nodes No. 51-65 and No. 106- 120.
