The Cloning and Haplotype Analysis of MAP30 Promoter in Bitter Gourd

Abstract: In order to explore the expression regulation of MAP30 gene,the upstream sequence,1 682 bp in length including start codon of MAP30 gene was cloned in bitter gourd (Y5).1 500 bp upstream of the transcription start site was selected for analysis based on PlantCARE promoter prediction tools.The results showed that in addition to containing the core elements of TATA-box and CAAT-box,the MAP30 promoter also contained light response elements,abscisic acid response element,heat stress response element,drought response element,salicylic acid response element etc.The prediction results suggested that MAP30 promoter might be involved in the induction of light, anaerobic, stress, abscisic acid, salicylic acid and ethylene, and it might also play important roles in the efficient expression in endosperm and circadian rhythm regulation. Take 30 bitter gourds germplasm as materials,the SNPs and InDels distribution in promoter region of MAP30 was analyzed.37 SNPs loci and 6 InDels were found.3 haplotypes were identified in MAP30 gene promoter through SNPs grouping.Y5, Y7, Y16, Y39, Y58, Y60, Y66, Y77, Y83, Y96, Y100, Y112 and Y121 belonged to group 1, Y43, Y50, Y90, Y108, Y113, Y115, Y140, Y141 and Y144 belonged to group 2, Y69, Y72, Y85, Y131, Y139, Y146, Y147 and Y153 belonged to group 3. Combining with the positions of cis regulatory elements, 5 SNPs, including T/C in 342 loci, A/G in 584 loci, G/T in 783 loci, A/G in 872 loci, T/G in 1087 loci, located in the cis regulatory elements region, the involving cis regulatory elements were MBSI, ABRE, ARE, 5UTR Py-rich stretch, GAG-motif and TC-rich repeats. The changes of the transcription factor binding sites caused by SNP may play important roles in regulating the expression of MAP30 gene. This study laid a theoretical foundation for further revealing the transcription regulation mechanism of MAP30 gene.
