Construction of DNA Fingerprinting and Analysis of Genetic Diversity with SSR Markers for SCRC Series Transgenic Bt Cotton Varieties

Abstract: SSR markers were used to analyze the fingerprinting and genetic diversity of 25 SCRC series transgenic Bt cotton varieties. Twenty-eight primers screened out from 412 SSR primers, with high polymorphisms and good repeatability, were regarded as core primers in the study. These primers whose SSR loci distributed on 21 chromosomes of cotton,have 2-8 alleles, with an average of 4.39; polymorphic information content (PIC) about 0. 63-0.93, with an average of 0. 84. These results suggest the core primers have preferable polymorphism. The digital fingerprintmg was used to indicated all estimate the varietres genetrc could be distinguished by 5 primer combinations at least. UPGMA cluster analysis distances and to construct a dendrogram by NTSYS 2. 1.The similarity coefficients of 14 SCRC conventional varieties were dance of genetic diversity. These results sources. between 0. 35-0. 87,with an average of 0.57,indicating relative abundance of genetic diversity. These results can provide technical support for cross breeding of SCRC germplasm resources.
