Construction of Rice T-DNA Insertion Population and Screening of Mutants

Abstract: It is a primary method to transgene and study functional genome for agrobacterium tumefaciens-mediated.Rice cultivar Nipponbare was transformed by this means,and 1833 independent T-DNA insertion population were obtained.T0 transgene plants were testified by PCR,GUS and Southern blot,and 20 grain of T1 seeds were planted to screen mutants in Hainan. 208 mutants were detected by observing all growth stages phenotype and inoculating experiment,and the percent of mutants was 11. 3 %,and the phenotype of mutants involved with plant height,leaf,growth stage,glume,glume color,resistance to disease and so on.Not only did the mutants provide excellent agronomic traits and new gene resource for rice breeding,but they were helpful to the study of rice functional genome.
