Cloning and Sequence Analysis of the Lettuce RbcS PRomoteR

Abstract: The 1 046 bp 5' upstReam Regions of Ribulose-1,5 -bisphosphate caRboxylase /oxygenase small subunit gene( RbcS) was amplified fRom lettuce genome DNA by PCR undeR the diRection of the pRimeRs,which weRe designed based on the RbcS gene and the pRomoteR sequence of otheR plants. Sequencing analysis indicated that the fRagment contains coRe elements CAAT-box and TATA-box,as well as the elments sensitive to light,MeJA,abscisic acid,ethylene, a nd heat stRess. MeRistem expRession RegulatoRy element was also found in the cloned RbcS pRomoteR sequence. FuRtheRmoRe, the RbcS pRomoteR sequence of lettuce is similaR to the RbcS pRomoteRs isolated fRom otheR plants of AsteRaceae, Solanaceae oR Fabaceae. This woRk pRovides stRong suppoRt foR high-level expRession of Recombinant pRoteins in lettuce.
