Effect of Osmotic Stress on the Content of Conjugated PA to the Thylakoid Membrane of Maize Seedlings

Abstract: The changes in contents of noncovalently conjugated-polyamines (NCC-Pas) and perchloric acid insoluble covalently conjugated-polyamines (PISCC-Pas) to thylakoid membrane were investigated in seedling leaves of two maize (Zea mays L. ) cultivars, Nongda 108 (drought-tolerant) and Yedan 13 (drought-sensitive) under osmotic stress. On the 7th day after polyethylene glycol (PEG)-6000 treatment, the contents of NCC-spermidine (Spd), PISCC-putrescine (Put) and PISCC-Spd decreased more significantly in Yedan 13 than in Nongda 108. In Yedan 13, the contents of NCC-Put and NCC-spermine (Spm) decreased more significantly, while Nongda 108 showed unaltered levels in both of them. Under osmotic stress, exogenous Spd treatment inhibited the decrease of NCC-Spd level in Yedan 13 and enhanced its tolerance to stress. Treatment with ethylglyoxal-bis (guanylhydrazone) (MGBG), which is an inhibitor of Spd biosynthesis, enhanced the decrease of NCC-Spd level in Nongda 108 and decreased its tolerance to stress. Treatment with o-phen, an inhibitor of PISCC-PAs biosynthesis, enhanced the decrease of PISCC-PAs and decreased the tolerance of seedlings to stress. The results suggested that the stability of NCC-Spd, PISCC-Put and PISCC-Spd contents in thylakoid membrane could enhance the adaptation of maize seedlings to osmotic stress.
