Study on Propagation of Corynespora cassiicola in Cucumber

Abstract: Corynespora cassiicola is a more serious diseases in Cucumber(Cucumis sativus L.) throughout China and the world.This experiment as a fundamental research for cucumber resistance breeding and disease control was conducted to establish a more reliable sporulation method of corymespora cassiicola.The experiment was about the effect of twelve culture media composition,transfer method,hyphae colour lllumination condition and culture time on propagation of corynespora Cassiicola in cucumber.The results showed that a dark colony could produce more conidia than a light one.The fungi could sporulate a lot by spreading loopful of surface hyphae over CZA120g and CZA80g media,but not in culture fluid.Continuous light and dark treatment did not have any significant effect on the hyphae growth and dark condition was preferable for sporulation.The 5th day saw a peak for conidial formation at 28℃ in the dark,and the conidia concentration could reach to 20.7×105 spores/mL.
