Construction of Expression Vector with Glyphasate-resistant Gene and Its Genetic Transform in Maize

摘要: 以pCambia3301载体为基础,以玉米Ubiquitin(Ubi)启动子、拟南芥叶绿素转运肽(Chloroplast transit peptide,Ctp)基因、抗草甘膦Epsps基因为元件,构建了玉米高效双元表达载体,通过冻融法将重组质粒导入根癌农杆菌LBA4404,并通过农杆菌介导法转化玉米幼胚,获得了抗草甘膦无抗生素标记的转基因玉米植株.经田间初步检测转化玉米植株具有一定的草甘膦抗性.

Abstract: High efficiency binary expression vector was constructed based on pCambia3301 vector,modified by the components of maize ubiquitin promoter,transit peptide of Arabidopsis thaliana chlorophyll (Ctp) gene,and resistance to glyphosate Epsps gene. The recombinant plasmid was transferred into Agrobacterium tumefaciens LBA4404 by freeze-thawing method,then transformed into maize immature embryos. The plants resistant to glyphosate with non-antibiotic marker gene were obtained. The preliminary identification in the field demonstrated that the transgenic maize plants were capable of resisting glyphosate to some extent.
