农杆菌介导遗传转化敏感基因型小麦的筛选鉴定

Screening of Wheat Genotype Sensitive by Agrobacterium tumefaciens

摘要: 敏感型受体小麦品种鉴定的匮乏制约了农杆菌介导小麦的发展。以Gus基因检测及PCR检测为方法,对68个品种的成熟胚愈伤检测Gus基因表达,对8个品种的幼胚转化植株进行PCR检测,结果表明:受体材料扬麦10、师栾02-1、石家庄5号、周麦18、山东01-35、泰山008、Y9-63、邯郸3475经PCR检测,检测到目标基因,为农杆菌侵染敏感型受体材料;扬麦10、师栾02-1侵染后,Gus基因能够表达,且以恢复培养14 d的侵染效果最好,Gus基因表达的愈伤数量增加与蓝斑体积增大,可作为体系优化的受体材料。

Abstract: The development of Agrobacterium -mediated transformation of wheat are restricted because the short of wheat genotype sensitive. This paper uses the method of the transformation expression of gus gene and PCRmeasure, de - tected 68 different materials of mature embryos in transformation expression of gus and 8 different materials of plants which were transformed from immature embryos, the results showed that foreign genes can be detect in Yangmai10, Shilu - an02 -1, Shijiangzhuang5, Zhoumai18, Shangdong01 -35, Taishan008, Y9 -63and Handan3475, so they are sensitive geno - type materials and can be used for the receptor of the Agrobacterium infection; Gus genes were expressed in Yangmai10 and Shiluan02 -1, the best effects of infection were resuming training in 14 The embryos members of Gus gene expres - sion were increased, and the volume of locus caeruleus were augmented?So Yangmai10 and Shiluan02 -1 can be used for the materials of the grading -up system.
