小麦TaNADP-ME1基因重组植物表达载体构建及对水稻的遗传转化

Construction of Plant Overexpression Vector of Wheat TaNADP-ME1 Gene and Genetic Transformation into Rice

摘要: 小麦TaNADP-ME1基因对干旱、盐、低温等非生物胁迫作出响应。利用重组技术成功构建了TaNADP-ME1的植物表达载体pCAME1,农杆菌介导法成功转化水稻品种日本晴成熟胚诱导的愈伤组织,通过潮霉素筛选获得了抗性愈伤,分化和生根后获得转化植株,PCR鉴定获得20株转基因水稻苗。结果为进一步确定TaNADP-ME1的基因功能奠定了基础。

Abstract: The wheat TaNADP-ME1 gene was responsive to drought,salt and low temperature. In this research we successfully constructed the plant overexpression vector pCAME1 by recombinant technology,transformed the callus which was induced from the mature embryo of rice by agrobacterium-mediated method,acquired resistant callus by hygromycin and transformed plant by differentiation and regeneration on media. The 20 transgenic rice plants were acquired by PCR identification. This research laid a foundation for identification the function of TaNADP-ME1 gene.
