影响小麦K199不同外植体愈伤组织诱导的因素

摘要: 以小麦科农199(K199)的完整种子、离体成熟胚和幼叶作为外植体,探讨了取材时间、取材部位、激素浓度和培养方式对小麦离体培养的影响。结果表明,刨碎的离体成熟胚和叶片基段为小麦组织培养的良好起始材料,种子萌发4 h为最佳取材时间,2 mg/L 2,4-D更有利于获得II型愈伤,光培养比暗培养更适用于叶片愈伤培养。

Abstract: The explants used in the study are whole seed, detached mature embryo and leaf segment of wheat cultivar K199. The factors affecting the induction and differentiation of calli were studied. The results showed that mature embry opieces and basal segments of young leaves were the best explant sources for K199. Good callus with high regeneration fre??quency could be obtained from medium containing Kinetin(KT)3 mg/ L and 2, 4 D ichlorophenoxyacetic acid(2, 4 D)0.01 mg/ L.
