应用浸种法导入外源DNA转化烤烟遗传性状变异的初步研究

A Preliminary Study on Using Germination Seed Soaking Method to Induce the Genetic Characters in Flue-cured Tobacco

摘要: 针对严重威胁烤烟大田生产的赤星病病害,培育抗病品种。以烤烟品种Nc89为受体,具有选择标记性状的Cv87为供体,采用浸种法将供体总DNA导入受体,获得具有目的性状的变异材料。实验结果表明:该方法可有效转移株高、株型、叶色、叶面积、生育期及抗病性等性状,并获得了较高转化率(21.2%)。

Abstract: This research was aimed at cultivating disease-resistant varieties. In this experiment, the method of germination seed soaking was used to study the way of exogenous DNA introduc-tion into flue-cured tobacco with Cv87 as the donor and Nc89 as the receptor. By means of pheno-type, disease-resistance and isozyme test, disease-resistant variation materials were selected. The result showed that exogenous DNA introduction could effectively transfer the character of plant height, plant type, leaf area, growth period and disease resistance, meanwhile obtained higher transformation ratio (21.2%). Its operation rule was simple, and it was proved an effective method.
