利用STS标记检测我国小麦推广品种的抗穗发芽基因型

Determination of Resistance Genotypes with STS Markers in Current Wheat Cultivars

摘要: 利用已开发的与穗发芽抗性相关的STS标记Vp1A3和Vp1B3,对我国46份代表性小麦品种进行穗发芽抗性基因的综合筛选。结果表明:用Vp1A3共检测出13种Vp-1A等位基因,分别是Vp-1Aam、Vp-1Aan、Vp-1Aao、Vp-1Abm、Vp-1Abn、Vp-1Agm、Vp-1Agn、Vp-1Ahm、Vp-1Aho、Vp-1Aim、Vp-1Ain、Vp-1Aio、Vp-1Ajm,各占总数的17.4%,8.7%,2.2%,4.3%,4.3%,6.5%,2.2%,2.2%,2.2%,15.2%,28.3%,4.3%,2.2%;用Vp1B3共检测出3种Vp-1B的等位基因,分别为Vp-1Bc、Vp-1Ba、Vp-1Bb,各占总数的71.7%,17.4%,4.3%。通过Vp1A3和Vp1B3的综合筛选,在46份材料中共选出5份抗穗发芽基因型材料,分别是绵阳31、豫麦35、凤麦24、CA0489和山东928802,其Vp-1基因的单倍型分别为:Vp-1Agm和Vp-1Bc,Vp-1Agm和Vp-1Bc,Vp-1Agm和Vp-1Bc,Vp-1Aam和Vp-1Bb,Vp-1Abm和Vp-1Bb。利用Vp1A3和Vp1B3对小麦的穗发芽抗性基因进行综合筛选,提高了选择效率。

Abstract: In this study, two markers Vp1A3 and Vp1B3 associated with PHS tolerance were used to evaluate the 46 white-grained wheats, in order to select out the cultivar which had the strong capacity of resistance PHS tolerance,and amplify these resistance PHS cultivars in wheat production and breeding for resistance PHS. In this study,total thirteen Vp-1A haplotypes,which were Vp-1Aam,Vp-1Aan,Vp-1Aao,Vp-1Abm,Vp-1Abn,Vp-1Agm,Vp-1Agn,Vp-1Ahm,Vp-1Aho,Vp-1Aim,Vp-1Ain,Vp-1Aio and Vp-1Ajm with frequencies of 17.4%,8.7%,2.2%,4.3%,4.3%,6.5%,2.2%,2.2%,2.2%, 15.2%, 28.3%,4.3% and 2.2% respectively,were detected by STS marker Vp1A3,and three Vp-1B haplotypes,which were Vp-1Bc,Vp-1Ba and Vp-1Bb with frequencies of 71.7%,17.4%and 4.3%, respectively,were detected by STS marker Vp1B3. In addition, there were five cultivars were selected out,which were Mianyang 31,Yumai 35,Fengmai 24,CA0489 and Shandong 928802,having the gene about resistance PHS. The haplotypes of Vp-1 in these five cultivars,Mianyang 31,Yumai 35,Fengmai 24,CA0489 and Shandong 928802,were Vp-1Agm and Vp-1Bc,Vp-1Agm and Vp-1Bc,Vp-1Agm and Vp-1Bc,Vp-1Aam and Vp-1Bb,Vp-1Abm and Vp-1Bb, respectively. Therefore, selecting efficiency for seeds dormancy could be improved by using a combination of the STS markers Vp1A3 and Vp1B3.
