Effect of PyTPS on Salt Tolerance and Quality of Transgenic Offspring of Peanut

Abstract: In this experiment,to analyze the effects of PyTPS from Porphyra yezoensis on the salt tolerance and quality characteristics of seed of peanut,the recombinant expression vector pCAMBIA2300-PyTPS harboring PyTPS was transformed into peanut via pollen-tube pathway method.Transgenic plants of T0 generation were analyzed by PCR and the positive rate was 34.7%.Transgenic seeds of T2 generation were tested for salt tolerance in a 0.7% NaCl solution and for quality by near infrared spectroscopy,and the expression level of PyTPS gene in transgenic plants was analyzed by semi-quantity RT-PCR.These results suggested that the germination rate was substantially higher for the seeds from some transgenic plants than for seeds from the non-transgenic parent in the NaCl solution and transcription of PyTPS gene was detected in some transgenic plants.Significant changes were found in quality characteristics including protein content,oleic acid and linoleic acid content,oil content of some T2-generation transgenic seeds.In a word,transgenic seeds of peanut exhibited enhanced tolerance to salinity stress and significant variation in quality characteristics after ectopic expression of the PyTPS gene in peanut.
