The Effect of Enucleation Time and Method on the Enucleation Efficiency Using in vitro Matured Goat Oocytes

Abstract: Oocytes were stained with Hochest 33342 at the different time during in vitro maturing of goat oocytes, e first aim was to deterimine the location relationships between polybody and nucleus chromosome;the second aim was to compare the enucleation efficiency when using two different enuetetion methods:blind-aspiration and fluorescence guiding.The result showed that at the 18 20,22,24 h during in vitro maturing of goat oocytes,the rate of type A oocytes W88 72.7%,43.7%,27.5%,16.7%,type B ooeytes was 9.l,31.3%,47.8%,55.6%,type C oocytes was 18.2%,25%,24.5%,27.7%.When enucleating at the 18,20,22,24 h during in vitro maturing of goat oocytes,the enucleation efficiency were 69.l%,58.2,50.7%,45.8%for bhnd-aspiration method however,all the enucleation efficiency were 100%when fluorescence guiding method used.
