Abstract: In order to offer more theory of female reproductive performance, the expression of GDF-9 gene mRNA at the period of pregnaney bovine was detected. First, total RNA was isolated from the bovine ovaries, and the primers were designed according to the GenBank, cDNA was amplified with RT-PCR, then cloned into Excoli DH5α by using pGM-T vector. Sequence analysis showed that the cloned 264 bp cDNA and the deduced amino acid sequence shared 97% and 94% homology respectively with ovine GDF-9 cDNA. According to βactin as inner Criteria, then the mRNA of GDF-9 gene were examined with RT-PCR. The result indicated that GDF-9 expressed in the bovine ovaries, oviduct, uterus and kidney, and the largest is in ovaries.
