Identification of Industrial Strains for Glucoamylase Production and Gene Cloning of Glucoamylase

Abstract: Sequence of the rDNAS were isolated and sequenced from four industrial strains of glucoamylase production. Based on BLASTn analysis of ITS1 and their morphological features, four strains were identified as Aspergillus niger, A. ficuum, A. oryzae, and Cunninghamella bertholletiae. Phylogenetic tree constructed by MegAlign indicates that A. niger, A. ficuum and A. oryzae are closely related, while C. bertholletiae are away from the group. The glucoamylase genes were cloned from the former three strains, and finally these genes get secreted expression in Pichia pastoris.
