Cloning of 5’Upstream Regulation Sequence for a High Molecular Weight Glutenin Gene in Wheat Store Protein

Abstract: The specific primers were designed according to the published sequence of wheat glutenin gene.The upstream regulatory region of a high molecular weight glutenin 12 subunit gene was amplified from wheat cultivar NE7742 genomic DNA by polymerase chain reaction.Sequencing analysis showed the cloned fragment contained 917 nucleotides and which located in 498-922 bp region shared a homology of 97.9% with the sequence published on genbank.There are 9 bp differences when compared with the reported sequence.The putative TATA box was present at position 836 to 841 bp and three possible CAAT-like boxes in 693 bp,730 bp and 808 bp respectively.A putative E-box and two E-likE-boxes were found.It was speculated that the presence of E-box were in responsible of gene endosperm-specific expression.
