A New Rapid DNA Extraction Method from Single Maize Seedling

Abstract: In order to resolve the critical problem in maize purity identification using.DNA fingerprint,a new rapid DNA extraction method was developed for maize single seedling. This method would not need liquid nitrogen, mechanical grinding, centrifugation, no precipitation, SDS, CTAB,nor chloroform. The quality of DNA extracted using this method was suitable for SSR analysis. It cost about 30 minutes to extract DNA from each of 100 seedlings, and in a single day,one person can complete DNA isolation from more than 2 000 individuals. It resolved the critical technique problem of DNA fingerprint widely used in identification of maize seed purtty.
