RAPD Analysis of Cotton Verticillium Wilt Pathogens

Abstract: DNA extracted by CTAB method has longer fragment, higher purity and better stability. So it is suitable for the DNA extraction of cotton Verticillium wilt pathogen. The RAPD analysis has been performed with 26 cotton Verticillium strains in 40 primers (OPY, OPB), 12 primers amplified polymorphic DNA patterns. By the cluster analysis, 26 strains are divided into 2 clusters (A, B). V. alboatrum belong to cluster A, cluster B comprises the other Verticillium strains (25 strains); cluster B can be divided into 2 subclusters including Ba and Bb, subcluster Bb followed by 3 groups: Bb1, Bb2, Bb3. The group Bb3 is the largest group, including defoliating strains (T9, VD-8, V44) and majority of Verticillium dahliae strains. The result of RAPD analysis of 4 Anyang strains showed the correlation of pathogenicity of cotton Verticillium wilt pathogen with RAPD analysis. RAPD analysis of different single spores also proved that the genomic variation may be occur among different single spores from one species.
