Cloning of TB-RBP Gene and Expression in Testis in Yak and Yak-cattle

Abstract: The mechanism of male sterility of hybridization of yak with ordinary cattle of other species is one of the hot topics to researchers. Through the comparative analysis of the specific expression genes in yak and yak-cattle testis,it is expected to provide a genetic reference for the regulation mechanism of male sterility in yak-cattle. It was detected to cloning and expression of TB-RBP genes in testis of yak and yak-cattle by quantitative RT-PCR. The results showed that CDS sequence of TB-RBP gene was 873 bp in yak,and partial CDS sequence was 587 bp in yak-cattle. The phylogenetic tree showed that the coding region of the TB-RBP gene of different species was highly conserved and high genetic similarity. Protein function prediction showed that TB-RBP protein belonged to the translin binding protein family,which was played an important role in regulating biological processes such as spermatogenesis. TB-RBP mRNA expressed in these nine tissues. The difference expression of TB-RBP mRNA in yak was higher than yak-cattle(0.01 < P < 0.05). TB-RBP gene was key gene for normal sperm development,and the expression level was lower in testicular tissue of yak-cattle. It indicated that male infertility in yak-cattle was associated with abnormal spermatogenesis. The results establish the base of farther research in molecular level. It may be helpful to offering theory gist in compare research between yak and yak-cattle.
