Electrophoretic Analysis on Lactate Dehydrogenase(LDH) and Malate Dehydrogenase(MDH) in Five Different Tissues of Oxygymnocypris stewartii

Abstract: The purpose was to learn about the biochemical heredity characteristics and to enrich the research data about the germplasm resources of Oxygymnocypris stewartii. By preparing concentration gradient polyacrylamide and using the vertical polyacrylamide gel electrophoresis,lactate dehydrogenase(LDH) and malate dehydrogenase(MDH) from five tissues(heart,crystalline lens,muscle,liver and kidney) of Oxygymnocypris stewartii were preliminarily studied.The results showed that:the band numbers of LDH isozymes from heart,Crystalline lens,muscle,liver and kidney were 12-16,17-18,6-13,6-10 and 12-17 respectively,and MDH isozymes 3,4,3-5, 2 and 4-7.The band numbers of LDH isozymes expressed in anaerobic organs such as muscle and liver were less than that in non-anaerobic organs such as heart and kidney.MDH isozymes in Oxygymnocypris stewartii were divided into supernate type and mitochondria type.LDH isozymes and MDH showed tissue specificity.More bands of LDH isozymes in Oxygymnocypris stewartii might be related to the interaction between it's unique habitant and the variation of subunit A and subunit B.The results could provide theoretical basis for the conservation and utilization of Oxygymnocypris stewartii from the biochemical genetics level.
