Cloning and Analysis of RcDof Gene Associated with Castor Dwarfing

Abstract: In order to research the features and biological function of RcDof gene in dwarf castor,stem tip genome DNA was extracted in growth climacteric age.Primers were designed according to obtained Zinc finger protein gene fragment.Using RACE technology,the complete full-length open reading frame of 924 bp was cloned,encoding 307 amino acids,belonged to C2C2 zinc finger protein,predicted protein molecular weight was 34.020 9 kDa,isoelectric point was 9.23.The predicted secondary structure α helix accounted for 1.63%,β fold accounted for 1.30%,other non-random coil accounted 97.07%,belonged to outer release proteins.Subcellular localization showed that the RcDof gene located in the nucleus.The bioinformatics analysis of RcDof gene will lay the foundations for the further study on its action mechanism and function characteristics in the castor dwarfing process.
