Genetic Diversity of Curvularia lunata by UP-PCR and ISSR Analysis

Abstract: In order to understand the physiological differentiation, the genetic diversity and the relationship of Curvularia lunata in Liaoning Province and Anhui Province.Genetic diversity of 30 isolates of Curvularia lunata from the major corn producing areas of Liaoning Province and Anhui Province in China was analyzed by UP-PCR(Universally primed PCR) and ISSR(Internal simple sequence repeat).The results showed that 4 primers of UP-PCR, 12 primers of ISSR Genetic 27 and 76 polymorphic bands, accounted for 77.78% and 89.47% of total bands respectively.These molecular markers revealed the genetic diversity of the strains tested, and there was no significant correlation between variation and region .The correlation between UP-PCR and ISSR, the dendrogram based on UP-CR results revealed that the 30 isolates were clustered into 7 groups at the threshold of genetic similar coefficient of 0.662, while ISSR were clustered into 8 groups at the threshold of genetic similar coefficient of 0.681.The ISSR technique was suitable for the genetic diversity analysis of C. lunata in view of the level of stability, operability and polymorphism.
