Isolation,Identification and Molecular Characterization of a H9N2 Isolate from a Broiler Flock in High Antibody Level Against H9N2

Abstract: A newly H9N2 isolate named A/Chicken/Shandong/LY/08 was collected from a broiler flock in high antibody level against H9N2.The biological test showed that the isolate A/Chicken/Shandong/LY/08 exhibited the IVPI value with 0, and the isolate shared a lentogenic motif 335RSSR↓GL340 at the HA protein cleavage site,which indicated that A/Chicken/Shandong/LY/08 was lentogenic.The HA gene nucleotides ( amino acids) homologies are 95.2%-99.6%( 95.4%-99.5%) between the isolate A/Chicken/Shandong/LY/08 and the Shandong H9N2 isolates.The HA gene nucleotides( amino acids) homologies are 82.6%-91.6%( 87.9%-92.2%) between the isolate A/Chicken/Shandong/LY/08 and A/ck/Korea/ms96/96,A/Parakeet/chiba/1/97,A/turkey/Wisconsin/1/1966 H9N2 isolates.Phylogenetic analysis of HA gene showed that the A/Chicken/Shandong/LY/08 was similar to that of the H9N2 isolates in China.
