Improving Resistance of Japonica Variety Wuyunjing 7 to Rice Stripe Virus Disease by Molecular Marker- assisted Selection

Abstract: To develop new germplasms resistant to rice stripe disease(RSV),the molecular marker- assisted se- lection(MAS) strategy was applied to improve the resistance of Wuyunjing 7.As the donor parent of resistance gene Stv- bi,the japonica rice Kantou 194 with high quality was used to improve the RSV resistance of Wuyunjing 7 with high yield but susceptible to RSV by the breeding strategy of directional backcross.In continuous backcross and selfcross generations,the SCAR(Sequence characterized amplified region) marker tightly linked with Stv- b iwas used for MAS,by combining with the identifications of field resistance to RSV and agronomic traits analysis. And finally,resistance gene was successfully transferred into Wuyunjing 7. 14 improved rice lines carrying the disease- resistance gene were obtained from BC3 F3 generation by molecular marker selection.These lines were the same as and even better than donor parent in comprehensive characters,which retained the characteristics of high yield and enhanced obviously the resistance to rice stripe virus.Utilization of technology systems intergrating directional backcross and MAS,the predictability,accuracy and efficiency of breeding for disease resistance could be improved prominently. Meanwhile,these RSV- resistant rice lines,newly developed could provided important genetic resources for the future resistance breeding.
