Abstract:
Polyclonal antibody was developed against compound antigens prepared from three Fusarium gramineurum isolates (BDX3-2, BDX4-5, HwH4-5), the causal agent of wheat scab, and an immunoassay for qualitative and quantitative detection of the pathogen was established. The antibody showed the highest titre at 1:64000, and the greatest sensitivity at 0.005 μg antigen/mL in ELISA. High reactivity was observed with Fusarium spp. pathogens, whereas no cross-reactivity with other thirteen fungi tested in ELISA. Biomass of F. gramineurum in the infected wheat seeds was quantified by using the ELISA, and the amount of the pathogen primarily agreed with the disease severity assessed visually.
