Abstract: The contents of sixteen main free amino acids in root system of maize cv. Yedan 13 and cv. Danyu 13 were determined by High Performance Liquid Chromatography (HPLC) at booting stage, filling stage and kernel maturity stage. The results showed that the contents of free amino acids in different secondary roots were significant difference in the same variety, and there was a positive correlation among the contents of free amino acids in the same layer of different varieties. The sequence of total content of free amino acids in different secondary roots was ninth layer>eighth layer>sixth layer>seventh layer. The contents of glutamic acid (Glu), proline (Pro), alanine (Ala) and asparagic acid (Asp) in maize root system were higher than the others, and the contents of lysine (Lys) and methionine (Met) were the lowest.
