Polymorphism Analysis by Sequence-related Amplified Polymorphism Markers in Brassica napus L

Abstract: Polymorphism among 1141B,KenC1,32B and 32C were analyzed with SRAP(sequence-related amplified polymorphism) markers.There were 13-36 clear polymorphic combination bands per primer,and the average was 5.8.About 419 amplified fragments and 116 polymorphic fragments were detected 20 SRAP primer combinations.The percentage of polymorphic fragments was 28.23%.The number of amplified fragments and polymorphic fragments per primer combination were 20.95 and 5.8,respectively.The polymorphism analyzed by F2 individual of 1141B Ken C1 with 24 primer combinations showed that there were 75 polymorphic bands,the average was 3.12.The χ2 test for allelic genotype of all loci indicated that 1.33%of the SRAP markers distorted from the expectation 3:1 ratio(dominance markers) and 1:2:1(co-dominance markers).It seems that SRAP markers was a high efficiency marker system in the paper.It showed that SRAP were an effective,cheap and dependable marker for polymorphism analysis and linkage map construction in B.napus L.
