Optimization of Parameters Influencing Gene Delivery into Maize Calli by Gene Delivery System

Abstract: Using maize calli as recipient cell,transforming parameters of PDS 1000/He gene delivery system and self made gene gun have been studied through GUS transient expression.The most suitable delivery parameters for PDS 1000/He gene delivery system are 1350psi of pressure,25 In·Hg of vacuum degree and 6 cm shoot distance of microcarrier;and for self made gene gun are 0.6 MPa pressure,10 c shoot distance of microcarrier.
