Abstract: The method of three factors second degree saturation D-optimum design was adopted in this experiment. The changes of fat content and morphology in embryo were studied under different treatments of density, nitrogen application and sowing date. The results showed that, during embryo establishment the fresh weight, dry weight, volume increased progressively, speed of growth was fresh weight >volume>dry weight. The moisture content decreased progressively. When the density and nitrogen application increased or sowing date being postponed, the fresh and dry weight, volume were falling, but moisture content was growing in the embryo. The whole change trend of fat content showed a single Peak curve, the peak value was 35d after insermination. With density and nitrogen application being increased or sowing date being postponed, the fat content was slowly falling after the peak value, but increased at mature period.
