N~+离子束注入玉米自交系变异的生物学效应与分子标记分析

摘要: 采用不同能量和剂量的N+离子束,对郑58、昌7-2和K12玉米自交系的干种子进行处理,结果表明,发芽率、发芽势和农艺性状变异都有很大差异,筛选出的N+离子束注入玉米自交系最佳诱变能量30 KV和剂量7×1016N+/cm2。同时利用10对SSR引物对玉米自交系进行扩增,共检测到39个等位基因,每对引物可检测等位基因2~5个,平均3.9个。根据SSR分子标记聚类分析,诱变能量30 kV和剂量7×1016 N+/cm2是诱变效果最佳的。3个自交系中郑58对N+离子束诱导引起的变异最敏感,N+离子束注入玉米自交系变异的生物学效应与分子标记分析结果基本一致。

Abstract: Three groups of dry seeds of maize inbred lines,including Zheng58,Chang7-2 and K12 were implanted with different doses of low energy N+ ion beam.We analyzed the dates of germination percentage,germinating energy and agronomic characters,and find that to the energy 30 kV,7×1016 N+/cm2 was the best implanted dose.At the mean time,the cluster analysis was done by molecular markers with 10 pairs of SSR primers.39 alleles were detected,2-5 alleles per primer,average 3.9.Zheng58 of the three inbred lines implanted with N+ ion beam was the most sensitive.According to the cluster analysis,the best implanted dose was 7×1016 N+/cm2 to the energy 30 kV.The results of the two methods were almost consistent.
