摘要: 为了给水稻功能基因组学研究提供材料,利用农杆菌(Agrobacterium tumefaciens)介导转化法转化水稻品种日本晴(Oryza sative L.ssp.japonica cv.Nipponbare),共建立了1833株独立的水稻T-DNA插入群体.T0通过PCR、GUS染色和southem blot分析证明了再生植株为转基因植株.随机选取种植T1种子20粒,进行突变体筛选.通过对各个时期表型的观察和接种试验,共发现突变体208个,突变率为11.3%,表型涉及株高、叶片、穗型、生育期、颖花、颖壳着色、抗病性等.这些突变体不仅创造了具有优良农艺性状的水稻育种材料,为水稻育种提供新的基因资源,而且为水稻功能基因组研究打下坚实的基础.

Abstract: It is a primary method to transgene and study functional genome for agrobacterium tumefaciens-mediated.Rice cultivar Nipponbare was transformed by this means,and 1833 independent T-DNA insertion population were obtained.T0 transgene plants were testified by PCR,GUS and Southern blot,and 20 grain of T1 seeds were planted to screen mutants in Hainan. 208 mutants were detected by observing all growth stages phenotype and inoculating experiment,and the percent of mutants was 11. 3 %,and the phenotype of mutants involved with plant height,leaf,growth stage,glume,glume color,resistance to disease and so on.Not only did the mutants provide excellent agronomic traits and new gene resource for rice breeding,but they were helpful to the study of rice functional genome.
