提高辣椒离体再生不定芽伸长率的研究

摘要: 通过调整培养基中的几种生长调节剂种类及其浓度组合,以及不同添加物,诱导不定芽分化与植株再生,有效地提高了不定芽的伸长率.结果表明:选用辣椒9~11 d苗龄的带柄子叶在培养基MB+BA 5.0 mg/L+IAA 1.0mg/L+GA31.0 mg/L+蔗糖3%+琼脂6.5 g/L+椰乳5%+AgNO35.0 mg/L上培养,分化频率达97.8%;芽丛在MB+ZT 1.0 mg/L+IAA 1.0 mg/L+GA31.5 mg/L+椰乳5%+AgNO35.0 mg/L培养基上伸长率达76.0%;幼苗在MS+IAA0.1 mg/L+NAA 0.2 mg/L培养基上能正常生根,生根率最高100%,并成长为健壮的再生植株。

Abstract: An efficient procedure to improve elongation rate of adventitious bud in vitro regeneration of Pepper was obtained by adjusting concentration and combinations of several plant growth regulators and some supplements.The cotyledons with petiole of 9-11-day-old seeding were cultured on MB(MS mineral+Vitamin B5)+BA 5.0 mg/L+IAA 1.0 mg/L+GA3 1.5 mg/L+sucrose 3%+agar 6.5 g/L+CM(coconut milk) 5%+AgNO3 5.0 mg/L.And the differentiation rate is 97.8%,with 1-2 adventitious bud rosettes per explants.Adventitious bud rosettes were elongated on MB+ZT 1.0 mg/L+IAA 1.0 mg/L+GA3 1.5 mg/L+CM(coconut milk) 5%+AgNO3 5.0 mg/L+sucrose 3%+agar 6.5 g/L medium,and the utmost rate of elongation is 76.0%.Rooting of the regenerated shoots was achieved on MS+IAA 0.1 mg/L+NAA 0.2 mg/L medium,and the rooting rate is 100%.It grew up to be strong regenerated plant at last.
