重金属镉、镍、汞对大麦过氧化物酶同工酶的影响

Effects of Cadmium, Nickel and Mercury on the Peroxidase Isozymes of Hordeum vulgare

摘要: 使用聚丙烯酰胺凝胶垂直平板电泳法,研究了不同浓度(10-4~10-8mol/L)的Cd2+,Ni2+和Hg2+对大麦根尖细胞过氧化物酶同工酶的影响。结果表明,10-4mol/LCd2+和Hg2+处理24h均比对照增加1条酶带(Rf=0.36),但前者缺失了1条Rf值为0.80的酶带8。处理48h后,10-6mol/L的Hg2 + 处理组又增加1条Rf值为0.36的新酶带。其余浓度的Cd2+,Hg2+和3种浓度的Ni2+均可使同工酶强弱发生改变。Cd2+和Hg2+对过氧化酶的影响明显大于Ni2+。3种重金属对大麦过氧化物酶同工酶的影响与浓度和处理时间有关。

Abstract: By using vertical polyacrylamide gel electrophoresis, the effects of three heavy metals at different concentrations on the peroxidase isozymes of barley root cells were studied. The results indicated that after 24 h treatment with 10-4mol/LCd2+ and Hg2+, one additional band (Rf=0.36) increased for each treatment, but a band (Rf=0.80) was missing in the former comparison with the control. After the treatment with 10-6mol/L Hg2+ for 48h, the treatment group added another band (Rf=0.36). The activites of peroxidase isozymes were changed greatly at rest concentrations ofCd2+、Hg2+ and 10-4~10-8mol/L Ni2+ treatment groups. It suggested that Hg2+ andCd2+ have stronger effects on the peroxidase isozymes than Ni2+ and the degree of effects of heavy metals on the peroxidase isozymes is dependent on their concentrations and treatment durations.
