Isolation, Identification and Molecular Characterization for Fgene of Newcastle Disease Virus Isolated in Shandong

Abstract: Four Newcastle Disease Virus isolates named SD01-SD04 were collected from chicken in Shandong in 2011. The four strains were purified by plaque assay can form typical cytopathic. The Fgenes of the isolates were cloned and sequenced to study its molecular characterization and hereditary variation. The deduced amino acid se-quence of Fgene showed that these viruses shared a velogenic motif 112RRQKRF 117at the cleavage site which were coincident with biological index. Phylogenetic analysis showed that SD01-SD04 and the viruses derived from chicken andgoose in China belonged togenotype Ⅶ. For Fgene of the isolates,the homologies of the deduced amino acid se-quences of SD01-SD04 with the same time isolates in China were higher compared with traditional trains. There was a closergenetic relationship between the four isolates and epidemic strains in China than foreign epidemic strains.
