Genetic Analysis of Genes for Yellow Pigment Content in Shaanxi Wheat Cultivars and Lines

Abstract: Color is an important trait exerting significant influences on the appearance of wheat-based products. Phytoene synthase (Psy) gene is a critical gene influencing biosynthetic pathway of yellow pigment (YP) in bread wheat. The aims of this study were to detect the allelic variations of Psy-A1 and Psy-B1 genes in 98 wheat cultivars and lines grown mainly in Shaanxi province using the functional markers YP7A and YP7B developed from the sequence polymorphisms of Psy-A1 gene on chromosome 7A and Psy-B1 gene on chromosome 7B,and to identify the distribution and to discuss the relationship between allelic variations of Psy-A1 and Psy-B1 genes to evaluate the feasibility of the markers for selecting wheat cultivars and lines with either low or high YP content. YP7A. The co-dominant marker of Psy-A1 gene can amplify a 194-bp and a 231-bp fragment in the genotypes with Psy-A1a frequently for high YP content and Psy-A1b mostly for low YP content,r espectively; and YP7B-1,another co-dominant marker of Psy-B1 gene,can amplify a 151-bp PCR fragment in accessions with the medium YP content allele Psy-B1a,and a 156-bp fragment in lower YP content accessions with Psy-B1b; two dominant markers YP7B-2 (428 bp) and YP7B-3 (884 bp) were designed for accessions with Psy-B1c and Psy-B1d,respectively. Allele Psy-B1c was associated with high YP content,but the phenotypic effect of Psy-B1d was not determined due to the limited number of accessions. The detecting results showed that the frequency of the genotype Psy-A1aPsy-A1bPsy-B1aPsy-B1bPsy-B1c and Psy-B1d is 39. 8%, 60. 2%,24. 5%, 67. 4%,7 . 1% and 1. 0%,respectively. In summary ,cultivars and lines with genotypes Psy- A1b and Psy-B1b should be mainly selected for the improvement and introduction of flour color in Shaanxi wheat cultivars (lines) . The functional markers YP7A and YP7B of phytoene synthase genes ( Psy-A1 and Psy-B1) can be directly used for promoting the breeding efficiency.
